ABSTRACT

OBJECTIVE:To study the potential mechanism of yam protein (DOT) in the prevention and treatment of diabetes-induced erectile dysfunction (DIED). METHODS :DIED model was induced by high-glucose and high-fat diet and intraperitoneal injection of streptozotocin (40 mg/kg). The experiment was set up in the normal control group (normal saline ), model group (normal saline ),DOT low-dose ,medium-dose and high-dose groups (0.3,0.6,0.9 mg/kg),sildenafil group (positive control ,4.4 mg/kg),with 9 rats in each group. In the stage of successful establishment of diabetes model and initiation of inducing DIED ,rats in each group were given relevant solution intragastrically ,once a day ,for consecutive 11 weeks. Body weight,fasting plasma glucose (FPG),the times and rate of penile erection ,fasting insulin (FINS),insulin resistance index (IR),the contents of endothelial nitric oxide synthase (eNOS)and cyclic guanosine monophosphate (cGMP)in penile cavernous tissue were determined so as to evaluate the intervention effects of DOT on DIED model rats. High-glucose damaged mice cavernous endothelial cells (MCECs)model was induced by 30 mmol/L glucose for 48 h,and then give DOT 125,250,500 μg/mL. The cell viability was detected so as to evaluate the effects of DOT on high-glucose damaged MCECs model. RNA-Seq mail:xingxin0902@163.com technology was adopted to screen the differentially expressed genes between normal MCECs and high-glucose damaged MCECs,high-glucose damaged MCECs and MCECs treated with 250 μg/mL DOT. Gene ontology(GO)function enrichment analysis and KEGG pathway enrichme nt analysis were performed for differentially expressed genes. The common differentially expressed genes between 2 groups were analyzed ,and mRNA expressions of six key genes were validated. RESULTS :Different doses of DOT could reverse the reduction of body weight ,the increase of FINS and IR ,the reduction of the times and rate of penile erection ,the decrease of eNOS and cGMP contents in penile cavernous tissue of DIED model rats ;above indexes of DIED model rats were reversed significantly after treated with high-dose of DOT(P<0.05 or P<0.01). 125,250,500 μg/L DOT could significantly improve the activity of high-glucose damaged MCECs (P<0.05 or P<0.01). RNA-Seq technology showed that compared with normal MCECs ,a total of 48 differentially expressed genes were found in high-glucose damaged MCECs. Compared with high-glucose damaged MCECs ,a total of 779 differentially expressed genes were found in MCECs treated with DOT. The differentially expressed genes of 2 groups were mainly cellular process in biological process annotation ,cellular part in cell component annotation and binding molecular function in molecular function annotation ,which were mainly enriched in extracellular matrix receptor interaction pathway ,mismatch repair pathway , phosphatidylinositol 3 kinase-protein kinase B (PI3K-Akt)signal pathway and so on. Among differentially expressed genes of 2 groups,13 common differentially expressed genes such as Aldh1a1,Abcc5,Tac1 were found. DOT could significantly reverse the expression of the above common differentially expressed genes in high-glucose damaged MCECs. After validation ,DOT could significantly reverse the mRNA expression of TGF-β3,Txnip,Aldh1a1,Loxl1,Mt1 and Mt2 in high-glucose damaged MCECs. CONCLUSIONS:DOT could improve the symptom of DIED model rats ,the mechanism of which may be related to biological pathway of inhibiting fibrosis and reducing oxidative stress ,so as to improve the endothelial function of cavernous body.

ABSTRACT

The aim of this study is to o explore the diagnosis, treatment and prognosis of idiopathic subglottic stenosis (ISS) The clinical data of 15 patients with idiopathic subglottic stenosis treated in our department were analyzed retrospectively. The degree of stenosis was classified by the Cotton Airway grading system of Myer, with 8 cases of gradeⅡ, 4 cases of grade Ⅲ and 3 cases of grade Ⅳ. The time of follow-up of HTSS was 0.5-10 years. All 15 patients were successfully extubated without asphyxia, decannulation and wound nonunion. For patients with idiopathic subglottic stenosis in the non-progressive stage, active surgical treatment strategy should be adopted and treated individually. The prognosis is satisfactory.

ABSTRACT

OBJECTIVE:To study the improvement effects of total ginsenosides on the senescence of PC 12 cells induced by D-galactose and its mechanism. METHODS :Rat pheochromocytoma (PC12)cells were treated with D-galactose to establish cell senescence model. CCK- 8 method was used to screen the D-galactose modeling concentration and total ginsenosides concentration. Normal control group ,model group ,total ginsenosides low and high concentration groups were set up. Cell senescence ,cell apoptosis rate ,apoptotic cycle and mitochondrial membrane potential (MMP),cell adenosine triphosphate (ATP)and reactive oxygen species (ROS)levels in each group were detected. The expression of apoptosis related proteins [B lymphoma 2(Bcl-2)and its related egg X protein (Bax),cytochrome C (Cyt-C)] and oxidative damage related proteins [nuclear factor 2 related factor 2 (Nrf2),heme oxygenase 1(HO-1)] were detected. In addition ,positive drug group [ 5 mmol/L N-acetyl-L-cysteine(NAC)] and positive control group [ D-galactose+5 mmol/L NAC] were set up to compare the levels of oxidative damage related proteins. RESULTS:D-galactose could significantly inhibit the survival rate of PC 12 cells,with a critical concentration of 20 mg/mL. The total ginsenosides could significantly increase the survival rate of D-galactose induced senescent cells with a median effective concentration(EC50)of 65 μg/mL,and then the low and high concentrations of total ginsenosides were set at 55 and 65 μg/mL. Compared with normal control group ,the number of aging cells increased ,the apoptotic rate and percentage of G 1 phase were significantly increased i n model group. the percentage of S phase ,MMP and ATP contents ,the protein expression of Bcl- 2 and Cyt-C in mitochondria were decreased significantly ,whileROS content ,the protein expression of Bax ,Nrf2 and Cyt-C protein in endochylema were increased significantly (P<0.05 or P<0.01). Compared with model group ,the number of E-mail:sunqiao150509@163.com aging cells reduced ,the apoptosis rates and percentage of G 1 phase were significantly decreased in total ginsenosides low and high concentration groups ,the percentage of S phase ,the contents of MMP and ATP (except for low concentration group ),protein expression of Bcl- 2,Nrf2 and HO- 1 as well as protein expression of Cyt-C in mitochondria were increased significantly ;ROS level (except for low concentration group )and Bax protein as well as protein expression of Cyt-C were decreased significantly. The protein expression of Nrf 2 and HO- 1 were increased significantly in positive control group (P<0.05 or P<0.01), but it was lower than that of total ginsenosides groups . CONCLUSIONS:Total ginsenosides can improve D-galactose induced senescence of P 12 cells,the mechanism of which may be related to activating Nrf 2 antioxidant signal pathway to antagonize D-galactose induced oxidative stress and alleviating mitochondrial dysfunction.

ABSTRACT

Objective@#To assess the outcomes of partial cricotracheal resection (CTR) and extended cricotracheal resection (ECTR) for severe laryngotracheal stenosis.@*Methods@#From November 2009 to September 2017, 18 patients underwent CTR and ECTR at the Department of Otorhinolaryngology Head and Neck Surgery, Tangdu Hospital, Air Force Medical University for severe laryngotracheal stenosis were reviewed retrospectively. There were 12-male and 6-female patients, with the age ranged from 4 to 56 years (median 25 years). The causes were postintubation in 11 cases, cervical trauma in 4, idiopathic in 3. The stenosis located in subglottic and tracheal (n=12), glottic and subglottic and tracheal (n=3), subglottic (n=2), and glottic and subglottic (n=1). Two patients had concurrent unilateral vocal cord palsy.One patient had undergone previous endoscopic balloon dilation and 8 patients had previous laryngotracheal reconstruction. The stenosis was graded according to modified Myer-Cotton classification as follows: Ⅲb (n=1), Ⅲc(n=1), Ⅳa (n=2), Ⅳb (n=12), Ⅳc (n=2). The surgical outcomes and complications were recorded.@*Results@#Among 18 patients,11 of the 12 patients undergoing CTR were decannulated. Five of the 6 patients undergoing ECTR were decannulated. Resected airway length ranged from 1.5 to 4.0 cm (median 2.8 cm). Surgical complications included infection of incision wound in 2 cases, anastomotic granulation in 2, cervical subcutaneous emphysema in 1, aspiration in 1, and unilateral arytenoid prolapse in 1. No recurrent laryngeal nerve injury or tracheoesophageal fistula occurred. The median follow up was 11 months.@*Conclusions@#CTR is efficient for severe subglottic and upper tracheal stenosis while ECTR is efficient for subglottic stenosis extended to the glottis. Both procedures also provide a salvage therapy for patients with previous failed treatments.

ABSTRACT

OBJECTIVE:To optimi ze the ratio of four comp onents of Compound renshen jianti formulation (Panax ginseng , Dioscorea oppositifolia ,Lycium barbarum fruit,Alpinia oxyphylla ),and to investigate its anti-fatigue activity and acute toxicity. METHODS:The water extract of Compound renshen jianti formulation was prepared by water extraction ,concentration and decompression drying. By single factor tests ,using weight-bearing swimming time as index ,the effects of four factors were investigated,such as the amount of P. ginseng ,D. oppositifolia ,L. barbarum fruit,A. oxyphylla . On the basis of single factor tests,using comprehensive score of weight-bearing swimming time ,serum urea nitrogen content ,liver glycogen content and AUC of blood lactate after exercise as index ,the formulation was optimized by Box-Behnken response surface method. The mice was divided into blank control group (water),positive control group (Renshen hongjingtian capsules ,0.135 g/kg)and compound low-dose,medium-dose and high-dose groups [the optimal ratio of Compound renshen jianti formulation extract (called“optimal compound formulation ”for short )4.08,8.16,12.24 g/kg,by crude drug] ,intragastric administration of drug or distilled water 20 mL/kg,once a day ,for consecutive 30 d. The weight-bearing swimming time ,the contents of serum urea nitrogen ,liver glycogen and blood lactate AUC after exercise were used to optimize its anti-fatigue activity of optimal compound formulation. The comprehensive score was calculated based on the measured data of mice in the compound formulation middle-dose group , and the difference between it and the theoretical prediction value was compared. The mice were given optimal compound formulation intragastrically (total dose 16.00 g/kg, by extract). The general state , body mass change , toxic characteristics and death of mice were observed and recorded for 14 days. Median lethal dose (LD50)and maximum tolerated dose (MTD)were measured. RESULTS :The optimal formulation ratio of Compound renshen jianti formulation included that P. ginseng 1.5 g,D. oppositifolia 10 g,L. barbarum fruit 10 g,A. oxyphylla 3 g. Results of anti-fatigue activity validation test showed that the optimal compound formulation could significantly prolonged weight-bearing swimming time ,reduced serum content of urea nitrogen ,blood lactate content and its AUC (except for low-dose group ),while significantly increased the content of liver glycogen (P<0.05 or P<0.01). Average comprehensive score of medium-dose group was 96.95,which was only 0.06% different from the theoretical prediction value of 97.01. The results of acute toxicity test showed that there was no death in mice. The oral MTD of the optimal compound formulation was more than 15 g/kg,which was non-toxic. CONCLUSIONS :The optimal Compound renshen jianti formulation has effective anti-fatigue activity of mice ,and has no significant toxic effect.

OBJECTIVE:To study the effects of gin seng root extract on autophagy ,proliferation and cytokine of splenic lymphocyte of mice ,and to study its mechanism. METHODS :The splenic lymphocyte of mice were divided into blank control group,positive control group (concanavalin A ,5 μg/mL),different concentration of ginseng root extract groups (32,125,500 μg/mL,by lyophilized powder ). After 48 h of culture with the corresponding medicine ,acridine orange staining method was used to detect autophagy of splenic lymphocyte ;CCK-8 assay was used to detect the cell proliferation ;ELISA assay was used to determine the levels of IL- 4,IL-6 and TNF-α in cell culture;Western blotting method was used to detect the expression of LC 3B and Beclin- 1,as well as the phosphorylation of AMPK ,AKT and mTOR. RESULTS :Compared with blank control group ,32, 125,500 μg/mL ginseng root extract could increase the number of acidic autophagosomes in splenic lymphocytes of mice(P<0.05 or P<0.01);125,500 μg/mL Ginseng root extract could significantly enhance the survival rate of splenic lymphocytes,the levels of IL- 4,IL-6 and TNF-α and the transformation of LC3BⅠ to LC 3BⅡ,significantly increased the protein expression of Beclin- 1, quinacrine cationic liposomes for treating non-small cell @163.com lung cancer[J]. J Drug Target ,2015,23(3):232-243. the phosphorylation of AMPK protein ,and significantly reduced the phosphorylation level of AKT and mTOR proteins ,with statistical significance (P<0.05 or P<0.01). CONCLUSIONS :Ginseng root extract can induce autophagy of mice splenic lymphocytes,activate the activity of splenic lymphocytes ,regulate the secretion of cytokine by activating AMPK and inhibiting the activation of AKT which can inhibit the activity of mTOR ,so as to exert immune enhancement effect.

Objective@#To explore the application value of thyroid alar cartilage(TAC) in the laryngotracheal reconstruction of subglotticstenosis in the paediatric population.@*Methods@#Twelve patients(7 males,5 females; range from 2.3 to 12.0 years) with subglotticstenosis who had undergone laryngotracheal reconstruction procedures at our hospital fromSeptember 2016 to July 2018 were analyzed retrospectively.The degree of stenosis was classified according to Myer-Cotton classification system:grade Ⅱ(n=4),grade Ⅲ(n=6) and grade Ⅳ(n=2). The stenosis planes were subglotticbut did not exceed the 3rd tracheal cartilage ring.@*Results@#Of all 12 patients, 11 were decannulated and 1 failed. All patients with grade Ⅱ and Ⅲ patients were decannulated at one procedure. There were no severe complications such as tube dislogement,asphyxia and unhealed wound.@*Conclusions@#The TAC for widening laryngotracheal lumen is relatively simple and reliable for laryngotracheal reconstruction in the pediatric population with subglottic stenosis. This would′t cause any structural damage to the laryngeal cavity.

Panax ginseng is a precious medicinal herb both in China and abroad with high medicinal and economic value.Ginseng disease has been recognized as the main factor restricting its application.Modern molecular biology for the disease resistance of ginseng promoted the development of new methods.In this study,it was found that the antimicrobial proteins of ginseng involved lipid transfer protein,cyclophilin,defensins,PR-4 and PR-10,showing inhibitory effects on various pathogens.These findings provided a reference for the control of ginseng diseases.

ABSTRACT

OBJECTIVE:To establish a method for the content determination of verbascoside in Naosaitong honeyed pill. METHODS:HPLC was performed on the column of Zorbax ODX with mobile phase of acetonitrile-0.1% phosphoric acid(16∶84, V/V)at a flow rate of 1.0 ml/min,the detection wavelength was 334 nm,column temperature was 30 ℃,and the injection volume was 10 μl. RESULTS:The linear range of verbascoside was 0.200-1.000 μg(r=0.999 8);RSDs of precision,stability and repro-ducibility tests were lower than 1%;recovery was 95.16%-99.78%(RSD=1.80,n=6). CONCLUSIONS:The method is simple and accurate,and suitable for the content determination of verbascoside in Naosaitong honeyed pill.

ABSTRACT

Objective:To establish the quality standard for Chidan Ganxian granules. Methods:TLC was used to identify Paeoni-ae Radix Rubra, Rhizoma Polygoni Cuspidati, Radix Salviae Miltiorrhizae, Herba Saururi and Radix et Rhizoma Rhei in Chidan Ganx-ian granules, and the content of paeoniflorin was determined by HPLC. The stationary phase was an Apollo C18 column ( 150 mm × 4. 6 mm, 5 μm), the mobile phase was acetonitrile-water(12. 5∶87. 5), the flow rate was 1. 0 ml·min-1, the detection wavelength was at 230 nm, and the temperature was 30℃. Results:The characteristic spots of the granules were the same as those of the standard samples without any interference from the negative control. Paeoniflorin had a good linear relationship within the range of 0. 120-1. 436 μg(r=0. 999 4), and the average recovery was 99. 8% with RSD of 1. 80%(n=6). Conclusion:The method is simple, ac-curate, reliable and reproducible, which can be used in the quality control of Chidan Ganxian granules.

BACKGROUND:Long-distance, large-range tracheal stenosis or defects are often seen in clinic, and laryngotracheal reconstruction is stil a difficult problem. OBJECTIVE:To investigate the effects of animal autologous cartilage transplantation in laryngotracheal reconstruction. METHODS:The cricoid cartilage and partial anterior tracheal wal from New Zealand rabbits were resected to prepare laryngotracheal defect models. Then, autologous costal perichondrium was taken for transplantation. After 8-24 weeks, surviving animals were sacrificed to observe the repair effects. RESULTS AND CONCLUSION:General observation showed that animals breathed and ate normaly, the implanted cartilage bonded tightly with the surrounding tissue, the wound healed wel without granulation tissue and scar formation, and there was a smooth inner surface covered by mucosa in the lumen. Under the light microscope, there was some mucosa generating at the wound site, and some fibroblasts and striated muscle cels existed in the outer layer, with a smal amount of new cartilage formation. There was also a linking between chondrocytes and muscle cels. These findings indicate that autologous cartilage transplantation can be applied for laryngotracheal reconstruction in animals, which has a good effect.

ABSTRACT

<p><b>OBJECTIVE</b>Retrospectively analyzed the influences of preoperative acoustic immittance pressure balance tests and intraoperative wet ear findings in myringoplasty surgery results.</p><p><b>METHODS</b>One hundred and twenty-one chronic suppurative otitis media patients underwent myringoplasty surgeries were included and divided into functional group and non-functional group according to preoperative acoustic immittance pressure balance tests. Meanwhile, cases were divided into dry ear group and wet ear group according to intraoperative findings. Postoperative hearing improvement and perforation rate were compared between the two groups.</p><p><b>RESULTS</b>Functional group had 72 cases, including 58 dry ears and 14 wet ears. Non-functional group had 46 cases, including 19 dry ears and 27 wet ears. Postoperative hearing improvement rate in functional group was 85.5% (59/69), and 72.1% (31/43) in non-functional group (χ(2) = 2.230, P = 0.093). Perforation rate in functional group was 16.7% (12/72), significant higher when compared with 2.2% (1/46) in non-functional group (χ(2) = 4.626, P = 0.015). Postoperative hearing improvement rate in dry ear group was 85.3% (64/75), and 71.8% (28/39) in wet ear group (χ(2) = 2.213, P = 0.085) . Perforation rate in dry ear group was 12.8% (10/78), and 7.0% (3/43) in wet ear group (χ(2) = 0.472, P = 0.377).</p><p><b>CONCLUSIONS</b>Non-functional result of preoperative acoustic immittance pressure balance test had significant relationship with the intraoperative wet ear findings. It may result in lower postoperative hearing level, and lower perforation rate. Large sample studies should be carried out in future.</p>

ABSTRACT

OBJECTIVE@#To learn the anatomical structure of the parapharyngeal space and to analyze the pathological type, preoperative evaluation and choice of surgical approach of parapharyngeal space tumors.@*METHOD@#To conduct a retrospective analysis of the clinical data of 43 patients who receive surgery in our department from March 2002 to January 2011. All patients undergo enhanced CT and MRI before surgery and enhanced CT reconstruction was also adopted in the cases after the year 2009. All 43 cases of surgeries were performed under general anesthesia. Of all the cases, the lateral neck path were chosen for 38 cases, lateral neck parotid path for 3 and lateral neck path combined with mandibular split operation for 2, 1 of which tracheotomy was conducted. Suction drainage was placed in the surgical cavity of all cases. And patients without contraindications were treated with glucocorticoids.@*RESULT@#In 42 of all 43 cases, the tumor was removed completely during the first operation while tumor reoccurred after 3 years and was cured by reoperation in the other one case.@*CONCLUSION@#Parapharyngeal space is deep and the anatomy structure is complicated. Cancer incidence rate is low and mostly benign in this regional. Surgical resection is the preferred treatment and lateral neck approach is most commonly used. CT and MRI is an effective preoperative examination and enhanced CT reconstruction can provide a more intuitive spatial conformation.

ABSTRACT

Objective To study the adsorption capability of chitosan(CTS)for Ni~(2+)in water.Methods The effects of different adsorption time(0-360 min),different temperature(25-60℃)and different initial pH value of the solution(4-8)on the adsorption were investigated.The adsorption kinetics and thermodynamics of CTS for Ni~(2+)in water were also studied and the IR of CTS and CTS-Ni~(2+)were determined.Results The maximal adsorption quantity could reach to 42.15 mg/g in the optimum condition that the temperature is 25℃,pH value was 7.0 and the adsorption time was 4 h.The adsorption kinetics showed that the pseudo-second-order equation was suitable for describing the adsorption of CTS for Ni~(2+)and the related correlation coefficients were more than 0.99.The calculated value of equilibrium adsorption quantity of Ni~(2+)was 45.05 mg/g.Study of adsorption thermodynamics showed that the linear regression equation of lnk and 1/T was lnk=-0.827 7/T+3.416 2,and the correlation coefficient was 0.998 9.The enthalpy change(△H~0)was 6.88 kJ/mol and the entropy change(△S~0)was 28.40 J/(mol?K).The IR showed that the coordination reaction occurred between-NH_2,-OH of CTS and Ni~(2+).Conclusion CTS can effectively remove Ni~(2+) in water.The adsorption of CTS for Ni~(2+)is spontaneous and endothermic.High temperature which is beneficial to the adsorption does not influence the adsorption quantity obviously.

ABSTRACT

Objective To observe the feasibility of using bone marrow-derived mesenchymal stem cells (MSCs) as seed cells for tissue-engineering in Otolaryngology. Method MSCs were isolated from bone marrow of human rib and purified by centrifuge and cultured in vitro. The proliferation and growth characteristics of MSCs were observed in primary and passage culture. Result Human bone marrow-derived MSCs showed active proliferation capacity in vitro in primary and passage cultures. Conclusion Human bone marrow-derived MSCs have relatively young biologic age and they can be used as the seed cells for tissue engineering.
